Strangely enough, I've always wondered about what Knights look like without their armor. Are they living or mechanical? Do they have a similar anatomy to humans? Who are they're species related to or derive from? Are they capable of vocal abilities?
The Knight's Species
We know they have to eat, since they have a chef who enjoys making killer cupcakes, but who knows?
At the very least, they'd have to be bio-mechanical. Their appearance and the flavor text on some weapons suggest this.
The knights are avatars controlled by people from another world. It explains why we have an overhead view of the knights and a HUD. The race controlling the avatars use satellite cameras to view their knights. The knights don't have a first person view and limited line of sight due to poor design, from which I can deduce that OOO are the manufacturers.

From the Scary Skelly Helm we know that Knights have hair on the backs of their necks (and that they have necks at all)
Also the Rock Jelly Core says that Knights have teeth.
there is some evidence that the Trojans are "hacked" Spiral Knights. Giving some weight to the idea that the Knights' must be machines.
On the other hand, when a Knight gets hurt, they can eat the beating heart of an enemy to recover... so bio mechanical?

I'm gonna stick with some sort of machines.
My theory is, SKs are in fact higher tier lifeforms. Their body unlike ours, are made out of mists ( energy solidifies into solid tissues ), Sk's body acts like that of a human, except that it is built out of mist rather than proteins. Adaptations such as small body can be used to avoid predators or get across the biomes easier ( heavy jungle in Isora? ) Large hands are adapted to make better tools ( all the weapons ) glowing eyes give SKs night vision( maybe Isora's night lasts way longer than that of earth(multiple moons) ). Larger Head indicates the superior growth of the brain, stumpy feet indicates... idk, either for combat, aka kicking to fight off enemies or fight for mates OR for travelling to hazardous terrian. The skill in alchemy shows that SKs are able to synthesis the primal energy around them and materialize them into actual matter (through a series of metabolic reaction (?)) This unique feature helped the SKs' ancestors make armors and weapons to secure their survival on Isora.
The reason why SKs left their homeplanet is probably due to a massive civil war, waged amonst the higher ranking vanguards, who lived off their middle rank loyalists And the low ranked kinghts leading by their revoulting middle ranking leaders, the reason for that is due to a serious inequality in terms of living standards. So, after many years, the civil war has finally ended ( vanguards won btw, "the rich grow richer" also applies on Isora ) The planet is devastated, infrastructures are heavily damaged and the planet can no longer support its population.
In desperation, the spiral order modified one of its last interspace mining ship "S.O.S. SKYLARK" (spiral order's ship= S.O.S.)into an interglatic spacecruiser, most of its ore silos are converted into rooms for the SKs . Then, well, u know the rest.
Note-i saw fanfic novel thread which described SKYLARK as a super space battlecruiser -.-|| really guys? Do u really expect a spaceship that carries MILLIONS of knights have spare space for nuclear rail guns or its powercells? no way...
They're wireframes. Look in the armour (accessory) display, and you'll see that they are made of floating blue lines.
I think that the knights are humans who wear bodysuits and "digital masks" that give them their glowing eyes (among other functions, like a HUD).
