In tier 1, all monsters do normal damage. so, we should focus on normal. but in tier 2 and 3 which type of defense should i focus on most?
normal > pierce, element, shadow or focus on those equally? What is the best combination for all round strategy?

Normal is most common (That's why it's called Normal)
Elemental is second-most common (Gun Puppies are everywhere, and arenas are full of Mecha Knights)
Piercing is third-most common (Every danger room until next patch will have either Slimes or Wolvers)
Shadow is least common (but definitely not "rare," still worth worrying about-- Never know when you might get stuck in a Graveyard, and Firestorm Citadel is almost all Zombies).

@Giga, now that Fiends inflict shadow, I'd say it's as frequent as piercing, especially since there are quite a lot of Fiend strata...
Build a set for all situations.
Specialize, do not generalize.
Focus on a set that defends against Elemental first, since it's the damage type you'll see the most often, then work slowly into Piercing or Shadow.
