Sure, you can use the buttons if you want (but obviously no pointing controls like on a Wii). You'd simply need to hook up the controller(s) to your CPU and get the proper drives to use them. If you need the drivers you can get them by simply searching for them online.

It's entirely possible to use your wiimote for pointing, but you do need the sensor bar set up.

Yes! I did this for a while when my Xbox 360 gamepad died. It was pretty complicated to set up but player surprisingly well for being a game not at all designed for it.

What you need to do is..

  1. Connect your WiiMote to the PC through bluetooth.
  2. Feed the digital input (buttons, d-pad) directly to keyboard/mouse button input through GlovePIE or FreePIE.
  3. Feed the sensor input to mouse position. To do this you need to run your Wii (U) while playing so that the sensor bar is powered. Note that the sensor bar is not actually a sensor, it just emits two points of infra-red light. Because of this, you can use a 3rd party wireless sensor bar which doesn't depend on the Wii (U) for power.
  4. Optionally, if you want full 360 degree movement instead of 8-way movement, you need to feed the Nunchuck's stick input to a virtual gamepad created through ppJoy or vJoy, which then give input to Spiral Knights.

Note that GlovePIE and ppJoy are both pretty old programs and neither are well supported on modern operating systems. However, I got them both working on Windows 7 64x. FreePIE and vJoy are both more modern open source alternatives, but they may be more buggy. I haven't used them for this purpose but I'm pretty sure it would work out if you just believe in yourself.

I literally just googled "how to use a wiimote on your computer for gaming", and this was one of the results, perhaps it will help? http://www.makeuseof.com/tag/how-to-connect-your-wiimote-to-your-pc/

Krakob basically broke down everything you need to know, but from the website, you should go to "Via Options > Preferences you can configure all of the options and button-mappings necessary to make the Wiimote your single-point user interface!", and from there just map everything you need to the correct keys.
