Right now i have Wolver Gear, Bristling Buckler, Night Blade, and Striker. I need to know what would be the best to level up first if i want to face the Royal Jelly. Some one said i should level up a sword first. Should i? And if so, should i get a vile striker first or a Silent Nightblade?

Level up the sword first(striker) then level up your shield since you'll die without some decent defense because even people with 5* gear die if they don't use their shield.
Ok, but should i really level up a sword to 4* star before i have 3* armor? And would a night blade be better overall because its better for The palace, and in the throne room it can easily rip up any jellies that annoy me or get in my way?
Being able to poison the boss is one of the most important things in the boss fight - far more important than shadow damage, which isn't increased as much against the boss as it is against regular slimes. As for the palace itself, slimes are easy to fight; you don't need a perfect sword for them, and you already have the nightblade.
Going from 3* to 4* isn't a massive damage boost in tier 2 (you probably won't get more than 10 additional damage per hit). The addition of poison to your sword, however, is very important for the boss.
As for armor, if you can reach the boss without upgrading it, you don't need it just yet; it won't help to upgrade it later, but 3* is more or less the cap on effectiveness in tier 2 for armor. I know you have 2* armor right now, but you pretty much just get one extra health per 200 energy if you upgrade it right now.
If you read enough of the threads on armor etc, you find a common theme. Having armor with the right resistances helps, but it's much better not to get hit in the first place. The safest thing to is to kill the monsters before they have the chance to damage you. The game rewards hit and run tactics rather than tanking.
Assuming that you're not intending solo jelly runs yet, Vile Striker is the way to go to help your party. It means that you have a better chance of poisoning RJ, which gives everyone a better chance of success. Using vials to poison him isn't always that successful... limited number of attempts and the other slimes often get in the way, also you can't guarantee that you will get any poison viles unless you join at tier 1 and buy them in Moorcroft
Vile Striker; it causes poison, which prevents the royal jelly from healing. This is equivalent to dealing an extra 57 damage per second to him for free, on top of the damage bonus moving to 4*.
Just note that it's not perfect elsewhere unless you get good with the playstyle.
